The advancement of technology in the scientific field has been very significant in recent years. As techniques improve, so does equipment. In the field of molecular biology, the PCR technique has been revolutionary.
PCR is a technique that allows the detection of organisms by amplifying a conserved region of their DNA. This process occurs at 3 different temperatures that repeat over several cycles:
- Denaturation: The temperature is increased to separate the DNA strands.
- Annealing: At this step, a temperature is established at which the primers attach to the DNA strands.
- Extension: At this temperature, the polymerase begins to extend the DNA strands, releasing the fluorophores that are excited and emit fluorescence.
The reaction takes place in an equipment called a thermocycler. It consists of a thermal block that changes temperatures to facilitate the reaction.
Real Time thermocyclers allow observing the results as the reaction occurs, as they measure the fluorescence emitted by the reaction and display it on a graph that is very easy to interpret.
